It wouldn’t be rude, depending on how you say it. You can’t exactly call her snotty to the boss’ face, but if you tell the boss perhaps that you feel as if you would be “more productive” on a shift without her, or as if “your personalities just don’t mesh” that might be a nicer way of saying how you really feel. Be careful since she’s the favorite!

Not at all, as long at your feelings towards her stay between you and your boss/person who makes the schedule. Once other people find out, rumors spread fast!

I think you should make it clear but also do it in a polite way. I can’t stand certain people sometimes too but oftentimes in life you just have to deal with it!
